干细胞全能性基因讨论 [复制链接]

各位,今年诱导成体细胞为iPS的干细胞全能性基因有哪些?哪几个效果更好,请大家讨论指教。我了解到的就是最开始的
0 \% \$ V4 j" i6 o/ ~0 q4 B* hklf-4:Krueppel-like factor 4 is a protein that in humans is encoded by the KLF4 gene. In embryonic stem cells (ESCs), KLF4 has been demonstrated to be a good indicator of stem-like capacity. It is suggested that the same is true in mesenchymal stem cells (MSCs)。

OOCT-4:Oct-4 (octamer-binding transcription factor 4) also known as POU5F1 (POU domain, class 5, transcription factor 1) is a protein that in humans is encoded by the POU5F1 gene.Oct-4 is a homeodomain transcription factor of the POU family. This protein is critically involved in the self-renewal of undifferentiated embryonic stem cells.As such, it is frequently used as a marker for undifferentiated cells. Oct-4 expression must be closely regulated; too much or too little will actually cause differentiation of the cells.& b  U3 z. z% ?' P# B8 v9 y0 r/ ~3 W
C-MYC:Myc (c-Myc) codes for a transcription factor that is located on chromosome 8 in humans and is believed to regulate expression of 15% of all genes [1] through binding on Enhancer Box sequences (E-boxes) and recruiting histone acetyltransferases (HATs). This means that in addition to its role as a classical transcription factor, Myc also functions to regulate global chromatin structure by regulating histone acetylation both in gene-rich regions and at sites far from any known gene.[2]
1 O* K$ ^# `( k9 s: g# U! w7 ? A mutated version of Myc is found in many cancers, which causes Myc to be persistently expressed. This leads to the unregulated expression of many genes some of which are involved in cell proliferation and results in the formation of cancer. A common translocation that involves Myc is t(8;14) is involved in the development of Burkitt's Lymphoma. A recent study demonstrated that temporary inhibition of Myc selectively kills mouse lung cancer cells, making it a potential cancer drug target。
0 B) J/ o6 f8 L+ u$ s" n- A4 FSOX-2:Sox2 is one of the key transcription factors required in induced pluripotent stem cells.[3] As forced Oct4 expression induces pluripotency in Sox2-null cells, a group of researchers concluded that the primary role of Sox2 in induced pluripotent stem cells is controlling Oct4 expression, and they perpetuate their own expression when expressed concurrently.[4]

后来还有lin-28:Lin-28 homolog A is a protein that in humans is encoded by the LIN28 gene.[1][2] It is a marker of undifferentiated human embryonic stem cells [3] and has been used to enhance the efficiency of the formation of induced pluripotent stem (iPS) cells from human fibroblasts. [4] It encodes a cytoplasmic mRNA-binding protein[5] that binds to and enhances the translation of the IGF-2 (Insulin-like growth factor 2) mRNA.[6]. Lin28 has also been shown to bind to the let-7 pre-miRNA and block production of the mature let-7 microRNA in mouse embryonic stem cells[7].

前四個是07年yamanaka和俞君英的兩篇文章中提到的吧?現在是不是用組蛋白的誘導更佳?

干细胞的MARKER基因太多了,除了以上的,FGF4,CRIPTO,FOXD3,CD9,LEFTY,LIFR,NOGGIN,PTEN,IMP-2,sFRP2等等,太多了
