Although embryonic stem cells currently enjoy the public limelight and show great promise
: ~' N- I* C9 h& M3 y; bfor cell based medical therapies, it is the adult stem cells which are responsible for the' T: I! C6 \0 Y4 H$ j9 `% P( g0 s" n I
body’s natural ability to fight disease, heal and recover, or fail and succumb to various) Z1 Y1 X+ o. F! n
maladies. The study of mammalian adult stem cells has surged recently, most likely from a
6 j" I. P: Z" T9 Qmaturation of stem cell studies in the classical developmental model organisms and in
" J4 G3 J) \4 @3 Q5 b4 y" ^hematopoeisis. All the tissues of the body examined so far are generated and regenerated
) @3 n1 g, @& Yfrom stem cells, it has been an important first step to adapt or devise new methods to
4 A2 x6 {6 V# z2 X0 C1 A; d' w9 [0 jidentify and obtain these cells in quantity and purity for further study. Culture techniques' q, q% W5 l( K
have been optimized for managing the growth and differentiation of stem cells in vitro; as- E- K; T p+ Y5 ]
some stem cells are pluripotent, often the method is to guide the fate of such cells among
7 q, L6 ?- g1 C4 |the possible differentiation fates. Much of this work, and that in the classical model organisms,
* g/ g4 F0 c( }9 k% ghas helped define the aspects of the stem cell environment or niche that are crucial" z+ D% }: t2 w, ~$ g2 [( Q
for both growth and differentiation, and these studies have moved in vivo at increasingly0 ~7 C, C3 p6 S/ L
higher resolution. Importantly, the in vivo niche is a current target for bioengineering the# p- j/ ]6 R, S4 ]' I
matrix and signaling factors.
! a2 g: f- m* wHerein, we present methods for studying six types of mammalian stem cells, mammary,
- Z6 Y2 L3 S1 V/ Tneural, mesenchymal, endothelial, dendritic, and muscle. We hope the approaches
3 j; k+ p, N' K9 }described in these chapters will assist you in obtaining, characterizing, and studying these! X) P* O! {: C1 }1 }3 g0 L g
cells or for adaptation to your favorite stem cells. One final note, should you find the
