STOCKHOLM, May 31 (Xinhua) -- Human embryonic stem cells have been cultured under chemically controlled conditions without the use of animal substances, which is essential for future clinical uses, according to a statement from Karolinska Institute in Stockholm on Monday.
) l+ }+ ?; D3 m0 m"Now, for the first time, we can produce large quantities of human embryonic stem cells in an environment that is completely chemically defined," said professor Karl Tryggvason, who led the study.
. ^' V  s* l, b' z( M"This opens up new opportunities for developing different types of cell which can then be tested for the treatment of disease," Tryggvason said.
) j' t: ]4 V+ F1 s! j. ~2 E% _A research team at Karolinska Institute has now managed to produce human stem cells entirely without the use of other cells or substances from animals. Instead they are cultured on a matrix of a single human protein: laminin-511, the statement said.
1 i( w+ o! z7 p; F! CLaminin-511 is part of the connective tissue and acts in the body as a matrix to which cells can attach, the statement said.
7 r4 p9 U5 G' ]( d3 B# R0 ]0 TThe researchers produced the laminin-511 using recombinant techniques.

We describe a system for culturing human embryonic stem (hES) cells and induced pluripotent stem (iPS) cells on a recombinant form of human laminin-511, a component of the natural hES cell niche. The system is devoid of animal products and feeder cells and contains only one undefined component, human albumin. The hES cells self-renewed with normal karyotype for at least 4 months (20 passages), after which the cells could produce teratomas containing cell lineages of all three germ layers. When plated on laminin-511 in small clumps, hES cells spread out in a monolayer, maintaining cellular homogeneity with approximately 97% OCT4-positive cells. Adhesion of hES cells was dependent on alpha6beta1 integrin. The use of homogeneous monolayer hES or iPS cell cultures provides more controllable conditions for the design of differentiation methods. This xeno-free and feeder-free system may be useful for the development of cell lineages for therapeutic purposes.
