Although embryonic stem cells currently enjoy the public limelight and show great promise
( ~! O" {1 Z5 Nfor cell based medical therapies, it is the adult stem cells which are responsible for the

maladies. The study of mammalian adult stem cells has surged recently, most likely from a/ w9 b, j2 |" O! j1 D
maturation of stem cell studies in the classical developmental model organisms and in
, r, T0 @, i. i* T5 v( ^2 h zhematopoeisis. All the tissues of the body examined so far are generated and regenerated
8 |( h# c2 r( K% k1 X( yfrom stem cells, it has been an important first step to adapt or devise new methods to
: ?) @" |, ?, J: V2 {identify and obtain these cells in quantity and purity for further study. Culture techniques/ j, i: E8 g, b3 {9 G3 o/ \! g
have been optimized for managing the growth and differentiation of stem cells in vitro; as
3 |( @/ A4 |4 m- E, c# X! L) E( xsome stem cells are pluripotent, often the method is to guide the fate of such cells among6 i) N* q$ m1 O$ o* j' D9 {9 t
the possible differentiation fates. Much of this work, and that in the classical model organisms,$ J+ }- e) b: z
has helped define the aspects of the stem cell environment or niche that are crucial
# c: s$ u3 g# H. u- V) t/ I y" ~for both growth and differentiation, and these studies have moved in vivo at increasingly1 h5 @+ t! V/ k0 L: C4 {9 d/ x
higher resolution. Importantly, the in vivo niche is a current target for bioengineering the* p7 R s- M# b* M8 }1 K) j
matrix and signaling factors.
3 I. k: O, ~% _* U1 wHerein, we present methods for studying six types of mammalian stem cells, mammary,
8 b" {( M- }- L, ~1 ~neural, mesenchymal, endothelial, dendritic, and muscle. We hope the approaches5 i# |: L m0 |) ^& W4 D
described in these chapters will assist you in obtaining, characterizing, and studying these
5 o% C: Z$ e- d4 acells or for adaptation to your favorite stem cells. One final note, should you find the
