Although embryonic stem cells currently enjoy the public limelight and show great promise
. K) l: Q, Q$ {9 Y afor cell based medical therapies, it is the adult stem cells which are responsible for the: `' G) G5 f* X) P! @0 s
body’s natural ability to fight disease, heal and recover, or fail and succumb to various y- A+ m! E5 Q* _9 M0 c [) ]/ Z# ]
maladies. The study of mammalian adult stem cells has surged recently, most likely from a
3 R2 o2 q; v6 hmaturation of stem cell studies in the classical developmental model organisms and in( ?6 I; j6 u4 ] Y" c9 s2 b2 h
hematopoeisis. All the tissues of the body examined so far are generated and regenerated$ c g; { X& Q
from stem cells, it has been an important first step to adapt or devise new methods to4 `4 a3 j; R3 r, a2 f4 B! _
identify and obtain these cells in quantity and purity for further study. Culture techniques& h0 d8 O, V5 q
have been optimized for managing the growth and differentiation of stem cells in vitro; as
$ |2 J/ y1 G+ U+ U( Ksome stem cells are pluripotent, often the method is to guide the fate of such cells among
7 @' b9 n4 C( Q/ ^4 y. X1 [the possible differentiation fates. Much of this work, and that in the classical model organisms,0 A6 w# D' E; u4 k
has helped define the aspects of the stem cell environment or niche that are crucial2 a7 a% G+ j' l) g: ?3 O8 ^( i7 U
for both growth and differentiation, and these studies have moved in vivo at increasingly
- \: ?, @1 I, g3 x# Ihigher resolution. Importantly, the in vivo niche is a current target for bioengineering the
* S/ t9 ~0 r. {( Jmatrix and signaling factors.7 Z- L# U, \ J$ A: s
Herein, we present methods for studying six types of mammalian stem cells, mammary,( n8 y. O, T( ?9 p2 B
neural, mesenchymal, endothelial, dendritic, and muscle. We hope the approaches
7 ^5 B& o+ g% L) M; F8 q6 p( Fdescribed in these chapters will assist you in obtaining, characterizing, and studying these& s5 {8 _, _! ^+ z$ T6 ?/ u- ~7 l
cells or for adaptation to your favorite stem cells. One final note, should you find the
